Interleukin 15 Receptor alpha (IL-15Rα) is a transmembrane glycoprotein that plays a pleiotropic role in immune development and function, including the positive maintenance of lymphocyte homeostasis. Fas Ligand, also known as FASLG and CD95L, is the ligand for FAS. It is a transmembrane protein which binds to TNFRSF6 FAS. ILDR2 is a member of the B7-like family of proteins that regulate T cell activity, is also a known endoplasmic reticulum molecule that regulates lipid homeostasis. SEMA4D is a member of the semaphorin family,contains one Ig-like C2-type domain, one PSI domain and one Sema domain. SEMA4D is strongly expressed in lymphoid tissues, especially in the thymus, as well as in the nervous tissues. However, SEMA4D is expressed at lower levels in testes, brain, kidney, small intestine, prostate, heart, placenta, lung and pancreas, but not in colon and liver. SEMA4D is a cell surface receptor for PLXN1B and PLXNB2 that plays an important role in cell-cell signaling. SEMA4D is involved in a number of fundamental biological processes such as promoting reorganization of the actin cytoskeleton, the migration of cerebellar granule cells and of endothelial cells and signaling via SRC and PTK2B PYK2, which then mediates activation of phosphatidylinositol 3-kinase and of the AKT1 signaling cascade. CD48 is a GPI-linked protein in the CD2 family of immunoglobulin superfamily proteins. CD48 is expressed on most lineage-committed hematopoietic cells but not on hematopoietic stem cells or multipotent hematopoietic progenitors. T-cell-specific surface glycoprotein CD28, contains an Ig-like V-type domain. CD28 is one of the proteins expressed on T cells that provide co-stimulatory signals, which are required for their activation. It is the receptor for CD80 and CD86. When activated by Toll-like receptor ligands, the CD80 expression is upregulated in antigen presenting cells (APCs). The CD86 expression on antigen presenting cells is constitutive. CD28 is the only B7 receptor constitutively expressed on naive T cells. It involved in T-cell activation, the induction of cell proliferation and cytokine production and promotion of T-cell survival.

Interleukin12 receptor subunit beta 1 (IL12RB1) is a type I transmembrane protein that belongs to the hemopoietin receptor superfamily. IL12RB1 can spontaneously form homodimers and -oligomers, which are able to bind IL12 with only low affinity. IL12 high affinity receptor complex is composed of two subunits designated IL12RB1 and IL12RB2. While IL12RB1 interacts with the IL-12p40 subunit, IL-12p35 is mainly connecting with IL12RB2. This receptor chain is also responsible for transmitting the IL12 signal into the cell. IL12RB1, to the contrary, is also part of the IL23R, where it interacts with the p40 subunit of IL23.
