产品描述 | C-DIM12 induced expression of Nurr1-regulated genes. C-DIM12 increased expression of transfected human Nurr1, induced Nurr1 protein expression in primary dopaminergic neurons and enhanced neuronal survival from exposure to 6-OHDA. |
体外活性 | C-DIM12在细胞系和原代神经元中诱导Nurr1和DA基因表达[3]。此化合物还能抑制体外星形胶质细胞的炎症信号。C-DIM12抑制脂多糖(LPS)诱导的BV-2微胶质细胞中NF-κB调控基因的表达,包括一氧化氮合酶(NOS2)、白细胞介素-6(IL-6)及趋化因子(C-C动机)配体2(CCL2),其效果通过Nurr1-RNA干扰而减弱。此外,C-DIM12降低了NF-κB-GFP(绿色荧光蛋白)报告细胞中NF-κB的激活,并增强了Nurr1在原代小胶质细胞核内的转移。C-DIM12减少了脂多糖诱导的p65与NOS2启动子的结合,并同时增强了Nurr1与p65结合位点的结合。C-DIM12还稳定了对抑制因子1沉默转录因子(CoREST)和核受体辅助抑制蛋白2(NCOR2)[2]的结合。 |
