Streptozocin is toxic to insulin-producing pancreatic islet B-cells and is commonly used in the construction of animal models of type 1 diabetes. 体外活性 | 方法:大鼠 β 胰岛素瘤细胞 Rin-5F 用 Streptozocin (1-10 mM) 处理 2-48 h,使用 MTT assay 检测细胞活力。 结果:在用 10 mM Streptozocin 处理 24 h 和 48 h 的细胞中观察到最大的抑制 (60-70%)。[1] 方法:神经干细胞用 Streptozocin (2.5 mM) 处理 2 天,使用 RT-qPCR 检测基因表达。 结果:Streptozocin 显著降低了 46.4% 的 GLUT3 mRNA 的相对表达水平,不影响 IR 和 GLUT1 的相对表达水平。[2] |
体内活性 | 方法:将 Streptozocin (40 mg/kg in the 50 mM sodium citrate buffer (pH 4.5),现配现用) 腹腔注射给 C57BL/6 或 CD-1 male 小鼠,每天一次,持续五天。给药期间提供正常食物和 10% 蔗糖水,第六天用普通水代替 10% 蔗糖水。 结果:重复低剂量 Streptozocin 诱导小鼠 1 型糖尿病。[3] 方法:将 Streptozocin (200 mg/kg in the 50 mM sodium citrate buffer (pH 4.5),现配现用) 单次腹腔注射给 C57BL/6 或 CD-1 male 小鼠。给药后提供正常食物和 10% 蔗糖水,第三天用普通水代替 10% 蔗糖水。 结果:单次高剂量 Streptozocin 诱导小鼠 1 型糖尿病。[3] |
细胞实验 | Streptozocin (STZ) stock solutions of 50 mg/mL in distilled Water (dWater) are freshly prepared for each experiment[2]. Human and murine cell lines are cultured in triplicate in 96-well plates at a density of 2×104 cells/well in the absence (untreated control) or presence of various concentrations of ALX (20-3000 μg/mL) or STZ (1-3000 μg/mL) for 48 h at 37°C under a humidified atmosphere containing 5% CO2. Cells incubated in complete media including dWater in a final concentration of 0.1% served as control for solvent toxicity and cells incubated in complete medium are used as a control for the experiments. The effects of the tested drugs on tumor cell growth or viability are determined employing the MTT assay in accordance with the manufacturer's instructions. The IC50values (drug concentration that induces 50% inhibition of the cell growth) are calculated using the GraphPad Prism 4 program[2]. |
