细胞焦亡,又称细胞炎性坏死,是一种程序性坏死;表现为细胞不断胀大直至细胞膜破裂,导致细胞内容物的释放进而激活强烈的炎症反应。按激活机制,可分为 Caspase-1依赖的经典途径和 Caspase-1不依赖(caspase-4/5/11)的非经典途径,caspase-1和 caspase-4/5/11通过切割 Gasdermin-D(GSDMD)蛋白,释放其 N 端结构域,该结构域结合膜脂并在细胞膜上打孔,导致细胞渗透压变化而发生胀大直至细胞膜破裂。两种途径都能同时诱导IL-1β和IL-18的前体进行切割,形成成熟的IL-1β和IL-18,并释放到胞外,造成炎症反应。细胞焦亡在感染性疾病、代谢疾病、神经系统疾病、心血管疾病、癌症等疾病中有广泛的研究,对细胞焦亡的研究有助于认识其在相关疾病的发生发展。 TargetMol 细胞焦亡化合物库收集了1066 种细胞焦亡相关的化合物,可以用于细胞焦亡相关研究。
